StepOne Software

StepOne Software from Applied Biosystems is a Windows-based application that controls StepOne and StepOnePlus Real-Time PCR Systems and analyzes qPCR data. It streamlines the workflow from experiment setup and plate layout to thermal cycling and results interpretation. Built-in wizards and templates help configure assays, define thermal profiles, assign samples, standards, and controls, and start runs with minimal setup time.

During data collection, the software provides real-time amplification plots and run status. Post-run analysis tools include automatic baseline and threshold settings, Ct determination, melt curve analysis for SYBR Green assays, and quantification options such as relative (ΔΔCt) and absolute (standard curve) analysis. StepOne Software supports both TaqMan and SYBR Green chemistries, reports quality metrics like efficiency and R2, and generates publication-ready charts and customizable reports. Data can be exported for further analysis or archiving, making it suitable for routine gene expression studies, pathogen detection, and other common qPCR applications on 48- and 96-well StepOne instruments.
